Some £25m of the cash will go to research on marine plastic pollution, while £20m will go on tackling plastic waste in developing countries.
Each of the countries will be asked to pledge action on plastic waste, from banning microbeads to committing to cutting down on single-use plastic bags.
Four Commonwealth countries have already joined the United Kingdom in the alliance - New Zealand, Sri Lanka, Vanuatu and Ghana.
She said: "The scourge of plastics is a global environmental challenge - and one that overwhelmingly impacts the livelihoods and health of the world's poorest people".
"As one of the most significant environmental challenges facing the world today, it is vital we tackle this issue so that future generations can enjoy a natural environment that is healthier than we now find it".
